Liraglutide with Pramlintide

Mice given olanzapine (Medak et al., 2024)

  • What was given — Male mice on standard chow got one injection of the antipsychotic olanzapine (5 mg/kg) with vehicle, liraglutide (25 µg/kg, a dose that did not protect on its own), pramlintide (1 µg per mouse) or both, and were followed for 2 hours; 4 to 6 mice per group (open-access full text).
  • What it found — Mice given both had a significantly smaller rise in blood sugar than mice given olanzapine alone, about equal to the two drugs’ separate effects added together. Glucagon fell below detection with the pair, and blood fatty acids fell with the pair but not with either drug alone. The authors: “There was an additive effect of pramlintide and liraglutide in protecting against olanzapine-induced hyperglycemia.”
  • Limits — One 2-hour experiment in male mice, 4 to 6 per group; no weight or long-term data. Funded by the Canadian Institutes of Health Research; the authors declare no competing interests.

A teenager in hospital with heart failure (Srivastava, 2023)

  • What was given — A 15-year-old with severe obesity (400 lb, BMI 71.6) in intensive care for heart failure was started on liraglutide (raised step by step to 3.0 mg a day) and empagliflozin. Pramlintide was also started in hospital but given only briefly: outpatient insurance would not cover it.
  • What it found — He lost 101 lb (25.2%) over 4 months, came off intravenous heart support and went home; his heart’s ejection fraction rose from 12–17% to 39%. After discharge he stayed on liraglutide and empagliflozin.
  • Limits — One patient; pramlintide was brief and given alongside liraglutide, empagliflozin and heart-failure care, so nothing can be credited to the pair. No funding; the author reports consulting fees from Novo Nordisk, Eli Lilly and Rhythm, outside this work.

No trial in people has tested pramlintide with liraglutide, and ClinicalTrials.gov lists none (searched October 3, 2026).
