Obese male rats, a comparison group (Petersen et al., 2026)

  • What was given — Male rats made obese on a high-fat, high-sucrose diet were dosed daily for 14 days with vehicle or one of three cagrilintide pairs at 5 + 5 nmol/kg: with tirzepatide, with semaglutide or with retatrutide; 10 rats per group. No group in this experiment got cagrilintide or tirzepatide alone (the paper’s figures and published source data).
  • What it found — Weight changed −10.6% on cagrilintide with tirzepatide, −13.1% with semaglutide and −17.6% with retatrutide, against +1.9% on vehicle (each pair vs vehicle, P < 0.0001). The tirzepatide pair lost less than the semaglutide pair (P = 0.0210) and the retatrutide pair (P < 0.0001). All three pairs cut food intake against vehicle (P < 0.0001).
  • Limits — One lab’s two-week study in male rats at one dose; without single-drug groups it cannot show whether the pair does more than either drug. Not blinded. Funding included Novo Nordisk Foundation grants; three authors co-founded a company developing obesity drugs, and some report speaking or counselling fees or research support from Novo Nordisk, Eli Lilly or Zealand Pharma.

No study in people has tested cagrilintide with tirzepatide. The three registered trials that name both compare CagriSema with tirzepatide; no one in them takes the two together (ClinicalTrials.gov, searched October 3, 2026).

Evidence: Approved drug. Zepbound: the weekly shot that beat semaglutide head-to-head. Main trial: 20.9% weight loss at 72 weeks.
